Ten years after the first American Pie movie, three new hapless virgins discover the Bible hidden in the school library at East Great Falls High. Unfortunately for them, the book is ruined, and with incomplete advice, the Bible leads them on a hilarious journey to lose their virginity.

After enrolling in her school, San Ho devotes his attention towards Chung, fueling the jealousy of Tse Tin, the son of the school dean. To make things even more complicated, Chung is infatuated with her physical education teacher (Russell Wong), and Daydreamer (Nadia Chan) has fallen in love with San Ho. Of course, what better way is there to resolve schoolyard romantic complications than a basketball game?
